OligoAnalyzer by Integrated DNA Technologies (IDT) is a powerful web-based tool for evaluating the thermodynamic and structural properties of oligonucleotide sequences before ordering. It calculates melting temperature (Tm), GC content, molecular weight, extinction coefficient, and nmol/OD values, and predicts secondary structures including hairpins, self-dimers, and hetero-dimers. Users can adjust hybridization parameters such as oligo concentration, Na⁺ concentration, Mg²⁺ concentration, and dNTP concentration for accurate predictions. It also integrates NCBI BLAST for sequence matching and provides a Tm mismatch analysis feature. It supports DNA, RNA, locked nucleic acids, and modified bases, making it indispensable for primer validation, probe design, and experimental troubleshooting.
